In vitro | This protocol, serving as a guideline adaptable to specific research needs, outlines the procedure for analyzing a sample's activity with 6-NBDG in an RPMI medium devoid of glucose. Begin by incubating the sample in RPMI containing 200 μM 6-NBDG. Subsequently, using curved forceps, immerse and evenly distribute the sample through a 20 μL drop of agarose (40°C) placed on a glass slide preheated to 40°C. Cover the sample with another preheated slide, ensuring the sample is fully encased within the agarose-6-NBDG solution. Finally, acquire confocal images using a confocal laser scanning microscope, with an excitation of 471 nm and emission ranging from 500–600 nm. |
